Adelaide had started the brighter and carved two opportunities in the first 10 minutes through Robert Cornthwaite and Richie Alagich.
But for the remainder of the first half chances were few and far between until Perth made the breakthrough on the stroke of half time.
Glory won a free-kick in a dangerous position just outside the box and Bertos stepped up to fire the ball into the back of the net.
Adelaide made a change at the break in an effort to improve their fortunes as Bobby Petta made way for Jason Spagnuolo.
And it was the substitute who had a hand in their first goal as he fed the ball to Cassio who in turn crossed for Djite and the Australia Under-23 striker struck the equaliser from close range in the 66th minute.
From then on Adelaide dominated the game and it was no surprise when they struck the winner seven minutes from time through Cassio.
Adelaide coach Aurelio Vidmar was thrilled with the result.
“The pleasing thing about this achievement is that they are working as a team," he said.
"It’s not about the individual, it’s about the team and that’s even with the players that aren’t in the squad and are on the bench.
“They can enjoy it but in two weeks time we will be playing for some serious points and that is what we’re working towards.”
Perth coach Ron Smith added: “The lads put everything they had into the match and that’s all you can ask of them.
“You couldn’t question their commitment at any stage today. There have been many encouraging signs throughout our pre-season campaign."
He added: “We all know there is plenty of hard work ahead and areas in which we can improve but the lads deserve a pat on the back for their efforts so far.
“I know they’ll use today’s result to spur them on and we’re all very much looking forward to playing at home and putting in a big performance in front of our members and supporters in round one."
